Social hierarchy is a structured ranking of individuals based on relative status, formed during initial encounters and shaped by subsequent social interactions. In animal studies, many researchers have focused on how social rank is shaped through group housing and investigated its underlying neural mechanisms. However, the effects of first contact on subsequent ranking formation remain relatively understudied. Here, we investigated the possible effects of initial encounters on the formation of social rank. C57BL/6J and stranger ICR mice (8-9-week-old, male) were subjected to a tube test. Then, the subordinate mice underwent a “WIN experience” using a forced-win tube test with a shutter placed behind the subordinate mice to prevent retreat. As a result, the subordinate mice repeatedly experienced pushing the dominant mice out of the tube (4 trials per day for 15 days). Finally, to assess the effects of the “WIN experience” on subsequent social rank, the tube test was conducted again with the same pairs as in the initial tube test and with novel pairs. Our preliminary data indicated that almost all ICR mice won C57BL/6J mice in the initial tube test. Unexpectedly, the subordinate C57BL/6J mice that underwent a “WIN experience” did not exhibit improved competitive outcomes over ICR mice, and consistently lost in subsequent tube tests. Since previous studies reported that “WIN experience” in subordinate mice can modulate subsequent social rank when assessed among C57BL/6J mice following group housing, our results suggest that the influence of “WIN experience” on social rank may be context-dependent, particularly with respect to social rank formed during first contact. In the future, we are going to increase the sample size and elucidate the underlying neural mechanisms.
